How do I reduce my thiol modified oligo?

1. Make a solution of 60 mM TCEP. (18 mgs TCEP in 1 mL H2O)
2. Add 125 uL 0.1M NaHPO4, 0.15M NaCl, pH 7.26, vortex & make certain oligo is in solution.
3. Add 75 uL 60 mM TCEP to oligo solution.
4. Vortex.
5. Let sit for 2 hrs at room temperature.

  1. Hi, I am looking for a dTTP-C6-SS-O-DMT where modification is done at 3′ and I wonder if you supply it?
Do you know if it serves as a substrate for DNA Polymerase I (Klenow)?
I have digested pMXB10 with HindIII (Linearized it) and now I want to fill the 5′ overhang and put the Sulfhydryl group at both 3′ ends; make a HS-C6-DNA-C6-SH structure.
    Best Regards

    • Dear Reza,

TriLink does not offer a 3’-C6-SS-O-DMT-5’-dTTP in our stocked catalog and I was unable to find a source for this material. If interested, you can request a Custom Chemistry Quote for the compound through our website. Additionally, we have not tested the incorporation of this compound with DNA Polymerase I. If you order the custom synthesis, we would recommend testing both DNA Polymerase I and Terminal Transferase. Please let me know if there is anything else I can help you with.

      Best regards,
      Elizabeth
